Purpose:To study molecular changes during structural healing in corneal stroma of cadaver eyes post Lasik and SMILE surgeries with quantitative proteomics. Methods:Cadaveric eyes(n=3)were treated for vision correction of -6D with ablation at 74-75µ and 65-66µ stromal depth for LASIK and SMILE respectively.Total protein from dissected stroma of each eye was extracted 14h post surgery and subjected to quantitative proteomic analysis by mass spectrometry. Results:Around 400 proteins were identified in stroma of LASIK and SMILE eyes.Cell cycle proteins (1433 class) and cytokeratins were higher in SMILE.Extracellular matrix (collagen 6 A1),cellular homeostasis proteins (Annexin A1, A5) were lower compared to LASIK.Lysozyme C, an inflammatory response protein was also lower in SMILE.Conclusion:Quantitative proteomic analysis of stroma post LASIK and SMILE hints at less inflammatory response, faster wound healing and stable extracellular matrix (ECM) in SMILE 14h post surgery compared to LASIK
